چکیده

Sigma receptors are unique endoplasmic reticulum proteins that mediate signaling for a variety of drugs. We determined the effect of sigma(1) receptor agonists on immune responses in a syngeneic lung cancer model. Sigma(1) receptor agonists, including cocaine, up-regulated splenocyte IL-10 mRNA and protein production in vitro in a sigma receptor-dependent, pertussis toxin-sensitive manner. In vivo, sigma(1) receptor agonists promoted tumor growth and induced IL-10 at the tumor site. Increased tumor growth was prevented by administration of specific Abs to IL-10 or by administration of specific sigma(1) receptor antagonists. We report that sigma(1) receptor ligands, including cocaine, augment tumor growth through an IL-10 dependent mechanism.

Acquired CD8 T Cell-Dependent Antitumor Regulatory T Cells and Uncovers Naturally Attenuates Their Ability To Induce Knockdown of HMGB1 in Tumor Cells

Although high mobility group box 1 (HMGB1) in tumor cells is involved in many aspects of tumor progression, its role in tumor immune suppression remains elusive. Host cell-derived IL-10 suppressed a naturally acquired CD8 T cell-dependent antitumor response. The suppressive activity of tumor-associated Foxp3 + CD4 + CD25 + regulatory T cells (Treg) was IL-10 dependent. Neutralizing HMGB1 impair...

Microenvironment and Immunology Immunotherapy with PI3K Inhibitor and Toll-Like Receptor Agonist Induces IFN-gþIL-17þ Polyfunctional T Cells That Mediate Rejection of Murine Tumors

The immunosuppressive microenvironment in tumors hampers the induction of antitumor immunity by vaccines or immunotherapies. Toll-like receptor (TLR) ligands have the potential to treat tumors, but they can exert amixture of positive and negative effects on inflammation in the tumormicroenvironment. In this study, we show that specific small molecule inhibitors of phosphoinositide 3-kinase (PI3...

Type II natural killer T cells foster the antitumor activity of CpG-oligodeoxynucleotides

Type II natural killer T (NKT) cells in cancer immunity are typically associated with suppression of tumor immunosurveillance through secretion of IL-13. We previously demonstrated that CpG oligonucleotide therapy activated Type II NKT cells to produce T helper type 1 (Th1) rather than T helper type 2 (Th2) cytokines. This cytokine skewing may manifest in Type II NKT cell antitumor properties i...

Immunotherapy with PI3K inhibitor and Toll-like receptor agonist induces IFN-γ+IL-17+ polyfunctional T cells that mediate rejection of murine tumors.

The immunosuppressive microenvironment in tumors hampers the induction of antitumor immunity by vaccines or immunotherapies. Toll-like receptor (TLR) ligands have the potential to treat tumors, but they can exert a mixture of positive and negative effects on inflammation in the tumor microenvironment. In this study, we show that specific small molecule inhibitors of phosphoinositide 3-kinase (P...

Microenvironment and Immunology IL-17A Produced by gd T Cells Promotes Tumor Growth in Hepatocellular Carcinoma

Interleukin (IL)-17A is expressed in the tumor microenvironment where it appears to contribute to tumor development, but its precise role in tumor immunity remains controversial. Here, we report mouse genetic evidence that IL-17A is critical for tumor growth. IL-17A–deficientmice exhibited reduced tumor growth, whereas systemic administration of recombinant mouse IL-17A promoted the growth of he...
